Neurobiotin image of an isolated (solitary) neuron cultured from rat cerebral cortex. The circular area, or island, was made by placing a collagen droplet on a thin agarose sheet. Astrocytes plated on such an island grow only within this area, which again restricts the growth of neurons that are subsequently plated. Under such conditions, all the synapses formed on this neuron are from this cell; conversely, all the synapses originating from this cell are formed on this cell. By observing spontaneous and stimulus-evoked EPSCs occurring at such autapses, some synapses are shown to be unable to release transmitters in a stimulus-evoked way, while spontaneously active.
